摘要:本课题以P123为软模板PC(聚碳酸酯膜)为硬模板利用微波辅助方法结合溶剂萃取的方法快速合成有机/无机复合介孔膜(HMSM)。将肌红蛋白通过振荡物理吸附到HMSM即Mb/HMSM,将Mb/HMSM通过Nafion溶液固定在玻碳电极表面,制成修饰电极Mb/HMSM/GCE。在最优条件下,修饰电极上的直接电化学行为表现为一对峰形良好的准可逆氧化还原峰,其形式电位为0.7 V。电极反应是一个表面控制过程。Mb/HMSM/GCE修饰电极对过氧化氢的电化学催化表明,Mb/HMSM/GCE能够电催化还原过氧化氢,而且过氧化氢浓度的线性响应范围是0.5到4.5 mM,催化灵敏度为2.43 µA mM-1 cm-2, 检出限为0.18 mM (S/N = 3)。本课题的实验结果虽不理想,但HMSM作为一个新型蛋白质载体,有望制备出长寿命,低检出限,更好应用前景的电化学生物传感器,并加深对于肌红蛋白电子传递过程的理解。23098

Development of functionalized Organic/Inorganic Hybrid Mesoporous Silica Membrane electrochemical sensors
Abstract:In this work, organic/inorganic hybrid mesoporous silica membranes (HMSMs) were rapidly synthesized using P123 as the soft template and PC (polycarbonate membrane) as the hard template by microwave-assisted method combined with solvent extraction. The myoglobin (Mb) was physically adsorded to HMSM by shaking to obtain Mb/HMSM, then the Mb/HMSM was fixed onto a glassy carbon electrode surface via Nafion solution, forming modified electrode Mb/HMSM/GCE. Under optimal conditions, the direct electrochemical behavior on modified electrode displayed a pair of well-defined quasi-reversible redox peaks, with the formal potential of ~0.7 V. Electrode reaction is a surface-controlled process. The electrocatalytic experiments of modified electrode towards hydrogen peroxide showed that, Mb/HMSM/GCE can electrocatalytically reduce hydrogen peroxide, giving a linear response range over 0.5-4.5 mM, catalytic sensitivity of 2.43 μA mM-1 cm-2 and  a detection limit of 0.18 mM (S/N = 3). These results show that though the performance of Mb/HMSM/GCE is undesirable, as a novel protein carrier, HMSM is expected to prepare a long-life, low detection limit, and more promising electrochemical biosensor, which will deepen the understanding of the myoglobin-related electron transfer process.
