中药黄芩生物转化初步研究。通过构建不同菌种的黄芩发酵转化体系,研究不同菌种的发酵对黄芩发酵液中总黄酮含量的影响以及是否能转化产生新物质

摘要:黄芩是我国应用广泛的传统中药材,在各种黄芩成分的研究中,黄芩的主要药用有效成分已确认为黄酮类化合物,生物转化是中药新药研制和开发的重要方法之一,而对于黄芩中黄酮的生物转化研究仍较少,本文旨在通过构建不同菌种的黄芩发酵转化体系,研究不同菌种的发酵对黄芩发酵液中总黄酮含量的影响以及是否能转化产生新物质。首先,构建黄芩生物转化体系:质量分数为5%的黄芩发酵液、发酵菌种为裂褶菌和茯苓菌;其次,对两种转化体系进行高效液相色谱测定,在裂褶菌-黄芩发酵体系中转化4天后发现明显新物质;最后,对两种转化体系进行了羟自由基(·OH)体外抗氧化活性测定,裂褶菌-黄芩发酵体系和茯苓菌-黄芩发酵体系的清除率分别达到了96。17%和83。04%。本文研究结果可为中药材黄芩的新药研制开发的进一步研究和利用打开一个新思路。94385

Abstract: Scutellaria baicalensis Georgi is a traditional Chinese medicinal materials widely used in our country, in the study of various composition of Scutellaria baicalensis Georgi, Scutellaria baicalensis Georgi, the main active ingredients of medicinal has confirmed as flavonoids, biological transformation is one of the important methods of the research and development of new drug of TCM, and for still less research medium yellow ketone bioconversion of scutellaria baicalensis, the purpose of this paper is to build different species of Scutellaria baicalensis Georgi fermentation by system, research on different strains of fermentation on the effect of Scutellaria baicalensis Georgi flavonoids content in the fermented liquid and can produce new substances。First, build the Scutellaria baicalensis Georgi bioconversion system: the mass fraction of 5% of Scutellaria baicalensis Georgi fermented liquid, to split pleat bacterium and poria cocos bacterium fermentation strains;Second, a high performance liquid chromatography (HPLC) determination of two kinds of transformation system, in the split fold bacteria - scutellaria fermentation system into four days later found significant new material;Finally, two kinds of transformation system for the hydroxyl free radical (· OH) in vitro antioxidant activity determination, split fold bacteria - scutellaria fermentation system and poria cocos bacteria - scutellaria fermentation system clearance rate reached 96。17% and 96。17% respectively。In this paper, the results for the further research of the new drug research and development of traditional Chinese medicine Scutellaria baicalensis Georgi and use to open a new way of thinking。
