枸杞中原花青素的组成与含量分析

摘要:本试验以宁夏黑果枸杞干品(2015年产和2016年产)为原料,对黑果枸杞中的多酚、黄酮、原花青素、花青素进行含量测定。通过大孔树脂对原花青素粗提取物进行纯化制备大孔树脂纯化样品,并以文生素C作为阳性对照,以DPPH自由基清除率、ABTS自由基清除率、超氧阴离子自由基清除率、FRAP值、总还原力等指标对黑果枸杞原花青素粗提取物和AB-8纯化样品的抗氧化活性进行评价分析,结果表明2015年产和2016年产的宁夏黑果枸杞浸提液中的多酚、黄酮、原花青素、花青素含量均较为丰富,且2016年产黑果枸杞中各化学成分含量均高于2015年产黑果枸杞,差异明显。黑果枸杞原花青素粗提物经AS-8树脂纯化后,原花青素含量由7.6%上升到76.8%。原花青素粗提物和纯化物均具有良好的抗氧化活性,原花青素纯化样品浓度达到200 ug/mL时对DPPH自由基、ASTS自由基和超氧阴离子自由基的清除率分别达到53.72%、95.36%和86.75%,抗氧化活性良好且各项抗氧化指标均高于原花青素粗提物,表明原花青素粗提物经过纯化后抗氧化活性得到提升,是潜在的具有良好抗氧化活性的天然植物成分。30675

Analysis on the Composition and Content of Proanthocyanidins in Chinese Wolfberry
Abstract:In this experiment, Ningxia Lycium ruthenicum dry goods (2015 production and 2016 production) as the raw material, we have determined the content of polyphenols, flavonoids, proanthocyanidins and anthocyanin in Lycium ruthenicum. Using the macroporous resin static adsorption experiments, we have determined the optimum resin type of extracting Procyanidins , the crude extract of procyanidins was purified to prepare macroporous resin purified samples. Taking vitamin C as a positive control, analysis the antioxidant activity of proanthocyanidins in Lycium ruthenicum crude extract and AB-8 purified samples ,with DPPH free radical clearance rate, ABTS free radical clearance rate, superoxide anion free radical clearance rate, FRAP value, total reducing power index. The results show that the content of polyphenols, flavonoids, procyanidins, anthocyaninsin 2015 production and 2016 production Ningxia Lycium ruthenicum extracts were abundant, and the content of chemical components in 2016 production Lycium ruthenicum were higher than 2015 production Lycium ruthenicum, and the difference is obvious. Purified with AS-8 resin, the content of proanthocyanidin in Lycium ruthenicum proanthocyanidin extracts increased from 7.6% to 76.8%. Procyanidins extracts and purified substance have good antioxidant activity, of which the proanthocyanidins purified samples at the concentration of 200 ug/mL ,DPPH free radical clearance rate, ASTS free radical clearance rate and superoxide anion free radicals clearance rate were 53.72%, 95.36% and 86.75%, good antioxidant activity and the antioxidant index were higher than that of proanthocyanidin extracts, showed that procyanidins crude extract after purification the antioxidant activity of it were promoted, which were potential of natural plant composition having good antioxidant activity.
