降解PAHs的海旋菌株LYG16的分离鉴定及其生长特性的研究

摘要采用富集培养方法,从连云港海域海水中筛选分离得到一株能以萘(naphthalene)作为唯一碳源生长的菌株,编号为LYG16,菌株细胞形态为杆状,长为27。0-28。0μm,宽为14。0-15。0μm,无鞭毛,没有运动性,革兰氏染色呈阴性;经过克隆后测序,得到1354bp的核苷酸序列,GenBank注册序列号为KT247808;利用EzTaxon Server和MEGA5。01软件对其进行同源性比较和系统发育学的分析,表明该菌株属于深海海旋菌属(Thalassospira Profundimaris),并命名为Thalassospira Profundimaris LYG16;进一步结合生理生化反应实验,该菌株的生长温度范围为30℃~40℃,盐浓度范围为10~40g/L,在pH为7。0,生长温度为35℃生长状况最好;通过进一步绘制细菌一步生长曲线,显示在最适培养条件下,0~8h为延滞期,8~64h为对数生长期,64h~75h后进入稳定期,80h后下降期。75027

Isolation, Identification and Growth Characteristics of  a  Thalassospira Profundimaris Strain LYG16 with PAHs Degradation

Abstract This thesis reported the isolation and characterization a bacterial strain from Lianyungang sea area which named LYG16 that has the ability to use naphthalene as sole carbon source for growth。 LYG16 cells are gram-staining-negative and flagellated。 They are short rods, 14。0–15。0 μm in width and 27。0-28。0 μm in length。 On the basis of the results of the phylogenetic analysis of 16 sDNA, LYG16 showed high similarities to T。povalilytica Zumi_95(T), so the strain LYG16 was identified as Thalassospira Profundimaris that in GenBank sequence number is KT247808 Based on integration of physiological and biochemical tests, the optimal temperature and pH of the strain are 30 ℃ and 7。0, the optimal NaCl concentration is 20g / L。 When strain LYG16 were under the conditions of 30°C 250 r/ min shaking culture growth curve, 0 h ~8 h was the growth lag phase,8 h~64 h was logarithmic phase, 64 h~75h was stable period and 80 h is the later period of decline。
