肝素钠废水中蛋白质降解菌筛选与鉴定

摘要:肝素钠废水中含有高浓度的无机盐以及大量的有机污染物,对环境具有潜在的威胁且难以处理。本论文开展了从肝素钠废水中筛选具有耐盐、高效蛋白质降解特性的菌株。通过奶粉培养基和废水培养基等限制性培养基方法,从淮安市双宝畜产有限公司下水道肝素钠废水中分离、筛选获得了一株耐盐且有蛋白质降解能力的菌株,通过菌落形态、生理生化特征将其初步鉴定为沙雷氏菌属。进一步研究分析了该菌的生理生化特性:在初始条件以为酪蛋白唯一氮源,3%盐浓度的发酵培养液中,该菌株在0到36h之间蛋白降解能力较为迅速,到32h时,该菌的蛋白降解率达到91.11%;在肝素钠废水培养基中,将该耐盐蛋白质降解菌在20 ℃,140 r/min的条件下培养72h,发现在肝素钠废水培养液中蛋白质降解率高达88.81%。通过本论文研究,对处理高盐废水中的蛋白质具有重要意义。65719

Abstract: Heparin sodium wastewater containing high concentration inorganic salt, and a large number of organic pollutants on the environment has the potential toxicity and hard to handle. This thesis conducted in waste water from heparin screening strains with resistance to salt, high protein degradation properties. Through milk powder culture medium method, and waste water medium to medium from huaian double treasure in livestock production co., LTD., sewer heparin sodium wastewater separation, screening for a plant salt tolerance and protein degradation ability of strain. By colony morphology, physiological and biochemical characteristics of the preliminary appraisal of serratia; Further research and analysis the physiological and biochemical characteristics of the bacteria: think casein is the only source of nitrogen in the initial conditions, 3% salt concentration in the fermentation broth, the strain between 0 to 36 h protein degradation ability is relatively quickly, to 32 h, the bacteria protein degradation rate of 91.11 percent; Medium in the heparin sodium wastewater, the salt tolerance of protein degradation bacteria at 20 degrees centigrade, 140 r/min under the conditions of train 72 h, found in water medium, heparin sodium protein degradation rate is as high as 88.81 percent. Through this thesis research, of the protein in high salt wastewater treatment is of great significance.
