摘   要随着现代生物技术的不断进步与发展,出现了越来越多的多肽类药物,多肽在临床医学中有巨大的药用价值。现如今已有100多种活性肽在人体中起作用,随着人们对生物活性肽的日益了解,对其在药理和医学上的应用产生了越来越浓厚的兴趣。分离纯化得到能用于疾病治疗的活性肽显得越来越重要。然而,从生物体内获取或基因通过基因重组技术从动物源得到的肽用于临床会产生免疫排斥反应。因此,肽类药物的合成是必须首要解决的问题。多肽样品纯度越高,对其在物化性质、生物活性及医药功能的研究越有益。89141

分离纯化多肽的方法有色谱电泳重结晶等多种方法,从现代分离科学理论得出,色谱和电泳是目前所知的应用最为广泛的方法,重结晶虽然是最有效的方法之一,但其应用主要限于小于5肽的纯化。目前,RP-HPLC可纯化50个左右残基的肽,而且分离度好,效率高,其在多肽的分离纯化和制备中的运用备受青睐。本文主要介绍多肽的一般合成方法和运用RP-HPLC技术的多肽进行分离纯化,分析影响条件,得到最优方法。

Abstract With the development of modern biotechnology, there are more and more polypeptide drugs, which have great medicinal value in clinical medicine。 Now there are more than 100 kinds of bioactive peptides play an important role in the central and peripheral nervous system, cardiovascular system, immune system and intestinal, with the increasing understanding of the mode of action of bioactive peptides, have become increasingly interested in its application in pharmacology and medicine。 It is becoming more and more important to isolate the bioactive peptides (endogenous substances) which have potential medicinal value and apply to the treatment of diseases。 However, it is difficult to obtain from living organisms, and peptides obtained from animal sources by recombinant techniques are used to produce immune rejection。 Therefore, the synthesis of peptide drugs is the first problem to be solved。 The higher the purity of peptide samples is, the more beneficial it is to study the physicochemical properties, biological activities and pharmaceutical functions。

Method for separation and purification of peptides with chromatographic electrophoresis recrystallization methods, derived from the theory of modern separation science, chromatography and electrophoresis is the most widely used method currently known, although recrystallization is one of the most effective method, but its application is limited to less than 5 of the purified peptide。 At present, RP-HPLC has the advantages of good separation effect, high resolution and high recovery rate, and can purify 50 peptides with left and right residues。 In this paper, the general methods of synthesis of peptides and the separation and purification of polypeptides by RP-HPLC technology were introduced。
