摘要:目的:本论文对采用毛细管电泳法法测定清开灵口服液中黄芩苷和绿原酸的含量进行了研究,为药用口服液中有效成分的定量研究以及中药成分和牛血清蛋白的相互作用研究提供实验方法和数据。建立关于中药中的绿原酸和黄芩苷的含量及其其它成分的方法分析。并研究中药成分与牛血清蛋白的相互作用。81277

方法:本实验采用毛细管区带电泳为方法,以及紫外检测模式,测定清开灵口服液中黄芩苷和绿原酸的含量,电泳条件: 35 mmol /L硼砂缓冲液,pH调节至8。5,未涂层弹性融硅毛细管(50μm×39。5 cm, 有效分离长度 34。8 cm)为分离通道,电动进样,分离电压为-18kv,温度为25℃,于326nm波长条件下对清开灵口服液中黄芩苷和绿原酸分离分析。结果:经过不断优化实验条件,黄芩苷和绿原酸两种成分在20min内与其他成分得到良好的分离。其中黄芩苷和绿原酸的峰面积和浓度,分别在0。1~0。5mg/ml,0。02~0。1mg/ml,的浓度范围内呈现良好的线性关系,其中检出限分别为0。04和0。02mg/ml,利用清开灵口服液做加标回收实验,黄芩苷和绿原酸的回收率均在96~102%之间。结论:本方法可靠有效,操作简便, 结果较为准确, 重现性好, 可用于清开灵口服液的质量控制。
