摘  要研究目的:利用四溴联苯醚(2,2',4,4'-tetrabromodiphenyl ether, BDE-47) 建立小鼠肝脏损伤模型, 研究曲克芦丁(Troxerutin)对BDE-47诱导的小鼠肝脏炎症损伤的影响及分子机制。方法:24只小鼠,随机等量分成4组:Control组、BDE-47组(150mg/kg/d)、BDE-47+Troxerutin组(150mg/kg/d)和Troxerutin 组(150mg/kg/d),连续喂养12周后处死,检测小鼠的体重、血清中ALT活力、ROS含量、NF-κB、F4/80和IL-1β表达量。结果:曲克芦丁改善BDE-47处理的小鼠血清中ALT的活性,能降低BDE-47处理的小鼠肝脏中ROS水平,能抑制BDE-47诱导的小鼠肝脏细胞NF-κB转核。曲克芦丁还降低BDE-47处理的小鼠肝脏中炎症相关蛋白F4/80、IL-1β的表达。结论:曲克芦丁抑制BDE-47诱导的小鼠肝脏炎症反应,其机制可能为曲克芦丁能够抑制BDE-47处理的小鼠肝脏组织中的ROS的浓度,进一步下调抑制细胞因子NF-κB的活性,从而减弱炎症相关因子F4/80、IL-1β的表达,保护了肝脏,从而为曲克芦丁的临床应用提供依据。70725

Troxerutin inhibits BDE-47-induced inflammation in mouse liver 

Abstract Objective: Mouse model of liver injury induced by 2,2 ', 4,4'-tetrabromodiphenyl ether (BDE-47) was established to explore the effects of troxerutin on inflammation in mouse liver and the molecular mechanisms underlying this action . Method: 24 mice were pided into 4 groups at random: control group , BDE-47 group(150mg / kg / d), BDE-47+ Troxerutin group (150mg / kg / d) and Troxerutin group (150mg / kg / d). Mice were killed after 12 weeks of treatment, and body weight, ALT activity, ROS content and protein expressions of NF-κB, F4/80 and IL-1β were measured. Result: Troxerutin improved ALT activity in the serum of mice treated with BDE-47, reduced the level of ROS in the livers of mice treated with BDE-47, and inhibited the nuclear translocation of NF-κB in the livers of BDE-47 treated mice. Ultimately, troxerutin suppressed the expression of inflammation related protein, such as F4/80 and IL-1β in the livers of BDE-47 treated mice. Conclusion: Troxerutin exhibited protective effects against BDE-47-induced liver inflammation by reducing the level of ROS, which thereby attenuated the nuclear translocation of NF-κB, and subsequent transcription of inflammation-related genes. This study indicates that troxerutin might be a candidate for the prevention and therapy of BDE-47-induced hepatotoxicity.
