褪黑激素对拟南芥根细胞钙离子运输的影响

摘要褪黑素是一类吲哚类激素,由脊椎动物的松果体分泌,主要参与动物昼夜节律的调节。现已证明褪黑素在高等植物中也广泛存在,但对其功能的研究仍不甚深入。Ca2+调控植物生长发育过程,作为第二信使,在信号转导方面具有重要作用。但是褪黑素是否能够诱导植物细胞钙信号产生仍不清楚,有待进一步研究。71131

本论文以拟南芥野生型Col-0和拟南芥钙通道蛋白突变体(膜联蛋白1[ANENXIN1]突变体:ann1和环化核苷酸激活钙通道2[CNGC2]:dnd1)为材料,利用非损伤微测技术研究了褪黑素对拟南芥根细胞Ca2+运输的影响。研究发现在瞬时褪黑素刺激试验后,野生型和突变型根细胞钙离子运输出现较大差别。施加一定浓度褪黑素处理后,结果显示野生型拟南芥根细胞瞬时钙内流增加,1-2分钟后恢复至处理前水平。而突变体ann1和dnd1根细胞钙离子流在褪黑素处理后无明显变化。根据实验结果,我们认为褪黑素能够明显诱导拟南芥根细胞外钙离子内流,引起细胞质钙浓度短暂升高,形成褪黑素特异性钙信号。此外,研究结果还显示Annexin1和CNGC2介导了褪黑素诱导的拟南芥根细胞外钙内流。研究结果为进一步探讨细胞质钙信号在褪黑素调控植物抗逆性中的作用奠定了坚实的基础。

The Effect of Melatonin on Ca2+ flux in Arabidopsis thaliana Root Cell

Abstract Melatonin is indole hormone produced by the pineal of vertebrate, mainly involved in regulating circadian rhythm of animals. It has been established that melatonin also found in higher plants, but still not research its function deeply. Ca2+ is involved in regulating plant growth and, as a second Messenger, plays an important role in signal transduction. 

In this study, wild-type Arabidopsis (Col-0), annexin1 mutant (ann1) and CNGC2 mutant (dnd1) were used to explore the role of melatonin on Ca2+ transport in Arabidopsis root cells. We used NMT System to measure effects of melatonin on Ca2+ of Arabidopsis thaliana root cell. Our results showed that a different Ca2+ flux responses were found among wild-type Arabidopsis and mutants. In Col-0 root cells, melatonin induced an instantaneous increase of Ca2+ influx and this Ca2+ influx maintained for 1-2 minutes and then recover to the control level. However, both ann1 and dnd1 mutants did not show the instantaneous increase of Ca2+ influx in response to 1 mM melatonin stimuli. Our results proved that melatonin can induce Ca2+ influx across the plasma membrane in plant cells and this process is possibly regulated by annexin1 and CNGC2 in Arabidopsis. Our data supplied important information for the further research on the mechanisms involved in the improvement of melatonin on plant stresses tolerance.
