摘 要:核糖体是细胞蛋白质合成场所,在植物生长发育和胁迫响应中发挥重要功能。RIO(Right open reading frame)家族属于非典型蛋白质激酶家族,其成员普遍存在于原核和真核生物中。在细菌、酵母和人类中的研究表明,RIO激酶参与核糖体亚基的加工成熟,而在高等植物中其功能尚不清楚。本研究采用生物信息学方法分别鉴定出水稻、玉米和拟南芥基因组中2个、3个和3个RIO成员。系统发育分析表明植物RIO属于RIO1和RIO2亚族,未发现RIO3亚族成员。蛋白质保守序列分析鉴定出10个保守基序,其中部分基序属于RIO催化结构域,其余基序的功能尚不明确。基因表达模式分析表明,植物RIO的表达均具有一定的组织特异性。水稻RIO植物激素和非生物胁迫处理下的表达分析显示,该家族基因被不同的植物激素处理显著诱导,并被各种非生物胁迫处理诱导或抑制。进一步采用基因组共表达分析进行功能预测表明,植物RIO激酶主要参与核糖体发生和生长发育过程。因此,RIO激酶在植物中可能具有保守的功能,即参与核糖体的发生,部分RIO可能参与植物激素和非生物胁迫响应。93563

Abstract: Ribosomes are the location where proteins are synthesized。 They play essential roles in plant growth and development as well as stress responses。 RIO (right open reading frame) family belongs to atypical protein kinase families, whose members widely exist in prokaryotes and eukaryotes。 Studies in bacteria, yeast and human beings have shown that RIO kinases participate in the processing and maturation of ribosome subunits。 Their roles in higher plants, however, remain illusive。 In this study, we identified 2, 3 and 3 RIO members from rice, maize and Arabidopsis genomes respectively using bioinformatics approaches。 A phylogenetic analysis showed that plant RIOs all belong to RIO1 and RIO2 subfamilies, whereas no RIO3 subfamily member was found。 Protein conserved sequence analysis identified 10 conserved motifs。 Some of them belong to the RIO domain, whereas the function of others were unknown。 Gene expression profile analysis revealed that plant RIO expression was tissue-specific。 Expression of rice RIOs under plant hormone and abiotic stress treatments showed that their expression was induced by different hormone treatments and was induced or inhibited by perse abiotic stress treatments。 Further, genome-wide coexpression analysis was perfromed to predict the putative function of plant RIO proteins。 This analysis suggests that plant RIOs might be involved in ribosome biogenesis and development process。 Thus, RIOs might have conserved function in plants, i。e。, participating in ribosome biogenesis。 Some of them were related to plant hormone and abiotic stress responses。
