丝胶蛋白水凝胶/支架辐照除菌技术的优化研究

摘要丝胶蛋白是构成蚕丝的两种主要蛋白(丝胶和丝素)之一。具有良好的生物活性和低免疫原性,已成为组织工程领域备受青睐的新兴生物材料。特别是2014年有关纯丝胶蛋白水凝胶的报道为丝胶蛋白在组织工程领域的应用提供了新的途径。然而有关丝胶蛋白的除菌技术仍缺乏系统研究。因此,丝胶蛋白高效除菌的研究十分必要。在本课题中,我们选用辐照除菌技术对不同阶段的样品(包括蚕茧、丝胶溶液、水凝胶以及冻干支架)进行处理,并对处理后的样品的理化性质进行系统地表征。86829

研究结果表明不同阶段的辐照处理对丝胶蛋白样品的某些性质产生了影响。SDS-PAGE凝胶电泳实验结果表明,经蚕茧和丝胶溶液的辐照灭菌处理,使其丝胶蛋白分子量显著减小;其中辐照后的蚕茧中,丝胶蛋白分子量变化较小,而辐照后的丝胶溶液中,蛋白分子量变化较大。FTIR实验是将经不同阶段辐照后的样品(包括蚕茧、丝胶溶液、水凝胶以及冻干支架)制成冻干支架,检测其蛋白质的二级结构,发现未处理支架中β-折叠含量为44。16%,经辐照处理的蚕茧中β-折叠含量为30。93%,经辐照处理的丝胶液中β-折叠含量为30。72%,经辐照处理的水凝胶中β-折叠含量为29。28%,经辐照处理的支架中β-折叠含量为39。38%。该数据表明辐照处理使丝胶蛋白中的β-折叠结构减少,从而使丝胶蛋白的水溶性增强[1],其中经辐照处理的支架中β-折叠含量变化较小。关于蚕茧溶解率的实验中,未处理蚕茧的溶解性为87。23%,经辐照处理的蚕茧溶解率为94。48%,证明辐照处理使蚕茧溶解率显著增大。另外,辐照对各阶段样品的支架孔隙率、膨胀性无显著影响。

Abstract Sericin protein are the two main types of silk protein (silk sericin and silk fibroin)。 Since possessing favourable biological activity and low immunogenicity, sericin protein become newly-developing and popular biomaterial material in the field of tissue engineering。 Especially, reports of sericin protein hydrogel provided the new application of sericin protein in the field of tissue engineering in 2014。 However, the sterilization technology of sericin protein is still lack of systematic study。 Therefore,it is necessary for the research of sericin protein effective antibacterial。 In this topic, we choose irradiation aseptic technique for different stages of the samples (including silkworm cocoon, sericin solution, water gel and freeze-dried scaffolds) for processing, and then characterized the physical and chemical properties of treated samples systematically。

The result shown that different stages of irradiation on sericin protein samples made different influence in protein properties。 SDS-PAGE gel electrophoresis experiment shown that sericin protein molecular weight decreased significantly after the cocoon and serici solution irradiation sterilization processing。 After irradiation of silkworm cocoon, sericin protein molecular weight changed small, while after irradiation of sericin solution, protein molecular weight changed a lot。 FTIR was mainly detected the protein secondary structure of the freeze-dried scaffold after different stage irradiation of the sample (including silkworm cocoon, sericin solution, water gel and freeze-dried scaffolds) processing。 The results shown that beta folding structure of the cocoon,s ericin protein in solution and water gel decreased, and the water soluble of sericin protein increased; while the protein secondary structure after the irradiation treatment shown no significant change。 After the irradiation treatment compared with untreated cocoon, cocoon its dissolution rate is significantly increased。 In addition, no obvious effect on stent porosity and swelling。after the irradiation samples at all stages。 丝胶蛋白水凝胶/支架辐照除菌技术的优化研究:http://www.youerw.com/wuli/lunwen_116805.html
