金线莲无网状叶脉与网状叶脉HPLC指纹图谱分析

摘要金线莲[Anoectochilus roxburghii(Wall。)Lindl。],即花叶开唇兰,别名金蚕、金线兰、金石松等,是兰科开唇兰属的一种多年生草本植物;同时也是我国传统的珍贵药材[3], 含有多种化学成分,包括黄酮类、多糖类、生物碱、挥发油、甾体、萜类等,具有抗肿瘤、降血糖、保肝、抗HBV、降血压等药理活性。平时所用的金线莲大多为有网状叶脉的表型,若无网状脉表型主要活性成分含量不亚于有网状脉表型,也可以对无网状脉表型进行利用。75225

HPLC指纹图谱法具有速度快、准确性高、稳定性高等特点,此方法作为一种综合的,可量化的鉴定手法被广泛应用于中药质量控制等各个方面。

金线莲苷作为金线莲的主要有效成分,有着良好的发展前景。而通过HPLC指纹图谱分析比较两种金线莲(无网状叶脉与网状叶脉)的主要有效成分,能进一步进行两种金线莲的质量优劣比较。

本实验准备了Agilent 1200型HPLC-MSD高效液相色谱仪、Agilent TC-C18(4。6×250,5m)色谱柱等器材,以甲醇与乙腈作为色谱纯,经过一系列的试验,最终确定了在色谱柱:Agilent TC-C18(4。6×250,5m);流动相:乙腈-甲醇-水溶液(85:10:5);检测波长:260nm;流速:1。0ml/min;室温的色谱条件下进行两种金线莲的HPLC指纹图谱分析。本次实验先对两种金线莲全草中的金线莲苷进行了细致的分离提取,分别配制相应的对照品溶液与供试品溶液,通过专属性试验、稳定性实验、重复性实验以及加样回收率实验验证了该方法高度的可行性,再由样品测定以及外标一点法计算出金线莲苷的含量以及平均含量。最终结果表明了该两种金线莲所含有的金线莲苷含量较为接近,证明了无网状叶脉的金线莲在一定程度上也可以作为常用的金线莲作为药材,而非冒牌的金线莲 ,这或许能够发展出一条崭新的获得金线莲以及相关成分的道路,使得金线莲的药用价值得到进一步的提升。

Abstract Anoectochilus formosanus [Anoectochilusroxburghii (Wall。) Lindl。], namely  Anoectochilus roxburghii,  Anoectochilus roxburghii,Jin Shisong etc。,which is a kind of perennial herb of the  Orchidaceae Anoectochilus; and also a valuable traditional Chinese medicinal materials, which contains a variety of chemical components, including flavonoids, polysaccharides, alkaloids, volatile oil, steroid, terpenoids, etc。, and also has the  Pharmacological activities of anti-tumor, decrease blood sugar, liver protection, HBV resistance, lowering blood pressure and others。The Anoectochilus formosanus which we usually used were mostly have the phenotype with reticulate veins, however ,if  the Anoectochilus formosanus without reticulate veins also have similar  active ingredient content with the other, we can also use the no stockwork phenotype。

HPLC fingerprint method has high speed, high accuracy, high stability, this method is a kind of comprehensive, quantifiable identification technique ,is widely used in many aspects, such as traditional Chinese medicine quality control。 

As the main effective components of Anoectochilus formosanus , kinsenoside has quite good development prospects 。By comparing HPLC fingerprint analysis of the main effective components of the two kinds of Anoectochilus formosanus (without reticulate veins and with reticular veins) , can further compare the quality of this two kinds of Anoectochilus formosanus。

This experiment prepared the Agilent 1200 type HPLC - MSD high performance liquid chromatograph, the Agilent TC - C18 (4。6 x 250 ㎜, 5 microns) chromatographic column and other equipments, with pure water, methanol and acetonitrile as chromatography, after a series of experiments, finally confirmed to use the chromatographic column(Agilent TC - C18 (4。6 x 250 ㎜, 5 microns)); Mobile phase( acetonitrile - methanol - water solution (90:5:5)); use the detection wavelength of 260 nm; the Flow rate of 1。0 ml/min and Room temperature。 under those chromatographic conditions, we can make the HPLC fingerprint analysis of these two kinds of Anoectochilus formosanus。 金线莲无网状叶脉与网状叶脉HPLC指纹图谱分析:http://www.youerw.com/shengwu/lunwen_86082.html
