摘要酸浆属植物是集食用、观赏和药用价值于一身的植物种质资源,为了实现其物种的快速鉴定、确保临床应用上的安全性和有效性,本研究采用DNA条形码序列psbA-trnH 对酸浆属植物进行鉴定研究,为酸浆属种质资源的保护和利用奠定基础。67301

方法:首先提取样本的基因组DNA,并进行电泳检测,然后通过PCR扩增获得扩增序列,所得序列使用CodonCode Aligner V3.7.1软件拼接,运用MEGA6.0软件和K2P模型进行多序列比对和遗传距离分析,从而构建NJ树,对psbA-trnH序列在酸浆属植物中的鉴定能力进行评估。结果显示,结合酸浆属植物K2P遗传距离的相关数据和种间种内较为明显的barcoding gap,可以明确地对种间种内植物进行区分。此次实验中检测序列的鉴定成功率时采用最近距离法(Nearest Distance)和相似性搜索算法(BLAST1),检测结果发现,上述两种方法针对psbA-trnH序列的鉴定成功率都非常高,因此能够凭借psbA-trnH序列对样本进行快速而准确的鉴定。系统发育树的建立则更为直观地体现了基于psbA-trnH序列的DNA条形码技术能够快速而准确的对酸浆属植物进行分子鉴别。结论:通过文献查阅及实验论证可知,psbA-trnH序列可以作为鉴定酸浆属植物的候选条形码序列,该DNA条形码技术在药用植物鉴别、濒危物种保护、种质资源评价等方面具有极大的应用前景和开发潜力,为将来的物种鉴别提供了新思路和新方法。

Abstract Genus Physalis is edible, ornamental and medicinal value of plant germplasm resources, in order to realize the rapid identification of the species, to ensure the safety and efficacy of clinical application. This study using DNA barcoding of psbA-trnH sequence on Physalis plants of the genus identification of, as Physalis genus germplasm resources protection and utilization lay the foundation. 

Method: First, genomic DNA sample, and electrophoresis, and then amplified by PCR amplification sequence, the sequence obtained use CodonCode Aligner V3.7.1 software stitching and use MEGA6.0 K2P model multiple sequence alignment and analysis of genetic distance to construct NJ tree of psbA-trnH sequence Physalis genus identification capability assessment. The results showed that psbA-trnH sequence have significant intra- and inter-specific pergences, and obvious barcoding gap, and high species discrimination rate (Based on both Nearest Distance and blast1 methods). Phylogenetic tree establishment is more directly reflected on psbA-trnH sequences of DNA barcoding can fast and accurate on Physalis plants of the genus molecular identification. In conclusion: psbA-trnH sequence can be as the identification of Physalis plants of the genus candidate barcode sequence, the DNA barcode technology in the identification of medicinal plants, protection of endangered species, germplasm evaluation has great application prospects and development potential, provides new ideas and new methods for species identification in the future.
