全氟辛酸对鲫鱼淋巴细胞抗氧化酶系统活性的影响研究

摘要有关全氟化合物(PFCs)环境污染行为的研究已成为环境化学的热点,因为全氟化合物(PFCs)是一类新型持久性有机污染物,具有环境持久性、生物富集性以及生物毒性效应。其中全氟辛烷磺酸(PFOS)和全氟辛酸(PFOA)是全氟有机化合物(PFCs)家族中两种代表性物质。全氟辛酸作为一种新型持久性环境污染物,对哺乳动物体内的抗氧化系统产生影响,进而对哺乳动物的生存造成影响。本研究以典型全氟化合物PFOA为目标污染物,选用藻食性鲤科鲫鱼(Carassius auratus)为实验材料,采用离体细胞体外诱导和暴露培养方法,利用酶标仪以及分光光度计对鲫鱼淋巴细胞的抗氧化系统指标进行测定,并且通过MTT法细胞增殖及细胞毒性检测法对细胞生长抑制率进行测定。我们将鲫鱼解剖后提取出肾脏,再从中提取出淋巴细胞,并将其暴露在1mg/L、5mg/L、15mg/L和30mg/L的PFOA中12小时,最后测定淋巴细胞的生长抑制率、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽转移酶(GST)和谷胱甘肽还原酶(GR),进一步说明全氟辛酸造成水生生物氧化损伤的影响机制。实验结果显示,随着PFOA暴露浓度的上升,SOD、CAT和GR的活性降低,GST活性增加,细胞生长抑制率逐渐增加,并呈现显著的剂量-效应。由此表明,高浓度的PFOA可诱导鲫鱼淋巴细胞抗氧化系统崩溃,造成淋巴细胞内产生氧化应激反应,脂质发生过氧化,进一步导致细胞发生凋亡。47528

Abstract

The studying of environmental pollution behavior of perfluorinated compounds (PFCs)   has become a hot topic in environmental chemistry, because perfluorinated compounds (PFCs) are new type of persistent organic pollutants, which showed environmental persistence, bioaccumulation and toxicity effects. The

 Perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) are two representative species of the perfluorinated compounds (PFCs). As a new type of persistent organic pollutant, perfluorooctanoic acid causes a series of influence to antioxidant defense system of mammals, also the survival of mammals. This study selected algal feeding cyprinid fish (Carassius auratus) as experimental materials, using typical perfluorooctanoic acid as the target pollutants, the training methods of induction and exposed cells out of bodies in vitro, the cells inhibition rates were measured by MTT proliferation and cytotoxicity assay,  index of oxidative stress in Carassius auratus lymphocytes were determined by enzyme mark instrument and optical photometric. We extracted kidney from carp, then extracted  lymphocytes from the kidney, which were exposed to 1mg/L, 5mg/L, 15mg/L and 30mg/L PFOA  12 hours, finally, determined the cell growth inhibition rate, activity of superoxide dismutase (SOD), catalase (CAT),glutathione reductase (GR) and glutathione

transferase (GST) to explains the mechanism of perfluorooctanoic acid  caused oxidative damage to aquatic

 organisms. Our results showed that with the increasing of PFOA exposed concentration, the activity of SOD, CAT and GR were decreased, while the activity of GST and cell growth inhibition rate were increased gradually,  showed dose effect in addition. This showed that the high concentration of PFOA can induce antioxidant system crashed in Carassius auratus lymphocyte, lead to oxidative stress, lipid peroxidation, Even apoptosis.
