大豆品种对大豆花叶病毒的抗病鉴定和标记辅助选择

摘要:本研究选择的268份大豆品种,均来自农业大学国家大豆改良中心的大豆种质资源,范围覆盖我国大豆优尔大生态区,人工接种我国黄淮海优势大豆花叶病毒株系SC3和SC7, 分别进行抗性鉴定和相关分析,进一步了解我国大豆品种对大豆花叶病毒病的抗病育种水平。同时利用与“科丰1号”和“齐黄1号”中SC3和SC7的抗性基因紧密连锁的分子标记进行抗病种质资源的分子标记辅助选择(Marker assisted selection, MAS)。实验结果显示:268份大豆品种对SC3和SC7表现抗病(高抗和抗)的品种百分比分别是42.38%和45.35%。中间类型(中抗和中感)的品种百分比分别是47.58%和29.00%。整体来看,占比例较低的是表现感病的品种,尤其是高感,都是6%以下。分子标记Satt558,BARCSOYSSR_02_0610,Satt266,Satt114和BARCSOYSSR_13_1136分别鉴定出与相应亲本特征谱带相同的种质资源分别有301、314、293、182和229份。其中,各标记基因型与大豆对SC3表型一致的种质资源分别有160、163、149、92和97份,符合率分别是53.16%、51.91%、50.85%、50.55%和42.36%;各标记带型与对SC7表型一致的大豆种质资源分别有160、172、138、89和103份,符合率分别是53.16%、54.78%、47.10%、48.90%和44.98%。本研究说明借助标记选择在一定程度上可以替代繁杂的人工接种鉴定,从而为加快大豆抗大豆花叶病毒病育种提供研究基础。31406

Identification of Resistance and Marker Assisted Selection of Soybean Mosaic Virus Resistance Gene Rsc-3 and Rsc7 in Soybean
Abstract:This study selected 268 soybean varieties with different types of soybean germplasm resources from the Agricultural University National Center for Soybean Improvement. These soybean varieties were inoculated with the soybean mosaic virus (SMV) strains SC-3 and SC-7 for identification of their resistance. Therefore, we can further understand the level of China soybean breeding. Also, SSR markers linked to SMV resistance genes Rsc-3 and Rsc-7 in Kefeng No.1 and Qihuang No.1 were used for Marker assisted selection (MAS) in order to find resistant germplasm resources. The results showed that among the 268 varieties, 47.58% are resistant to SC-3 and 45.35% are resistant to SC-7(including HR and R). Middle resistance levels (MR and MS) account for SC-3 and SC-7 are 47.58% and 29.00%, respectively. The proportion of S and HS were very low, especially HS which was even less than 6%. The proportion of SC3 resistant varieties was higher than that of varieties resistant to SC7. The MAS efficiency of SSR markers Satt558, BARCSOYSSR_02_0610, Satt226, Satt114 and BARCSOYSSR_13_1136 was evaluated in the soybean varieties. 301、314、293、182 and 229 varieties were identified for the same characteristic bands of corresponding parents. According to different SSR markers, 160 (53.16%), 163 (51.91%), 149 (50.85%), 92(50.55%) and 97(42.36%) are correct for resistant situation of SC-3; 160(53.16%), 172(54.78%), 138 (47.10%), 89 (48.90%) and 103 (44.98%) are correct for the resistant of SC-7. Therefore, the SSR markers can be used effectively in selecting for resistance genes Rsc-3 and Rsc-7 instead of inoculation identification for some situations.
