紫甘薯花青素抑制高脂诱导的肝脏内质网应激

摘要:本课题利用高脂饮食建立小鼠肝脏损伤模型,以探讨紫甘薯花青素(PSPC)抑制肝脏内质网应激的作用机制。将40只雄性ICR小鼠随机分成四组:正常对照组(Control)、HFD组、 HFD+PSPC、PSPC组。正常对照组及HFD组每日灌胃PSPC的溶剂,HFD+PSPC组及PSPC组每日灌胃PSPC(700毫克/千克/天),连续处理20周。处理20周后,测量并记录小鼠体重,处死小鼠,取肝脏组织及血清,制备肝脏组织匀浆。计算肝脏系数,测定ALT活性水平,取肝脏组织进行HE染色,以蛋白质免疫印迹法测定肝脏内质网应激相关蛋白水平和应激激活蛋白激酶水平。实验结果显示,相比于正常组的小鼠, HFD处理小鼠的体重、ALT活性及肝脏系数显著上升,肝脏细胞肿胀空泡化,肝脏内质网应激相关蛋白及应激激活蛋白激酶水平显著提高。HFD+PSPC组小鼠的体重、ALT活性及肝脏系数水平相比HFD组小鼠显著下降,内质网应激相关蛋白及应激激活蛋白激酶水平显著下降,与正常组小鼠无显著差异。由此得出结论,PSPC能够抑制高脂诱导的肝脏内质网应激。26273

 Purple Sweet Potato Color Restrains Hepatic Endoplasmic Reticulum Stress Caused by High Fat Diet in Mice
Abstract:
 Objective In this research, the liver injury mouse model induced by HFD (high fat diet) was employed to study the molecular mechanisms underlying the effects of PSPC on endoplasmic reticulum stress (ER stress). Methods  Male ICR mice were randomly pided into 4 groups: group Control, group HFD, group PSPC+HFD and group PSPC.Mice in group PSPC and group HFD + PSPC have daily oral administration of PSPC at doses of 700 mg/kg/day. After 20 weeks of treatment, mice were sacrificed. Liver tissue and serum were harvested to prepare the tissue homogenates. The wet weight of the liver, the activity of alanine aminotransferase (ALT), the histopathologic changes and the expression of related proteins were detected. Results The body weights, liver indexes and serum ALT activities were significantly increased in HFD-treated mice. Furthermore, HFD treatment induced obvious liver injury, including hepatocyte hypertrophy and vacuolization in mice. Ultimately, the protein expressions of ER stress related proteins and stress-activated protein kinases were markedly enhanced in the livers of HFD-treated mice. Interestingly, PSPC dramatically attenuated body weights, liver indexes, serum ALT activities, ER stress and protein expressions of stress-activated protein kinases in HFD-treated mice. Conclusion PSPC can effectively protect against HFD-induced liver injury by suppressing ER stress in mice.
