摘要:细菌的自杀行为是一种自然现象,并且这种行为是具有价值的。对这一现象的产生和文持及其对细菌群体进化的影响的研究在理论和实践上都具有重要意义。本课题基于原有的研究基础,以大肠杆菌top10F’为菌种,在LB液体培养基中培养,用IPTG作为诱导剂,使细菌发生群体感应自杀现象,比较细菌在不同生长条件下的生长曲线。IPTG作为诱发细菌自杀现象的外部诱导剂,在温度与pH值相同的条件下,IPTG的浓度越高,自杀行为越明显;AHL是细菌自杀行为中的重要因素之一,在温度较高,pH值较高的条件下AHL容易分解,稳定性降低。因此在IPTG浓度相同,pH值相同时,培养的温度越低,自杀行为越明显;而IPTG浓度相同,培养温度相同,培养基的PH值越低,自杀行为越明显。通过实验比较发现,在pH=5.7的LB液体培养基中加入浓度为1mM的IPTG,在30℃下培养的细菌产生最为明显的自杀现象。20477

The growth model of suicide population with quorum sensing in rich nutrient medium
Abstract: Suicide behavior of bacteria is a valuable natural phenomenon. To generate and maintain the appearance of the phenomenon and its effects on bacteria group evolution of the research is of great significance both in theory and in practice. This topic is based on the original research foundation, to top10F 'e. coli strains, in LB liquid medium cultivation, using IPTG as inducers, quorum sensing suicide phenomenon in the bacteria, comparing bacterial growth curve under different growth conditions. IPTG as external inducers of suicide phenomenon induced by bacteria, under the condition of temperature and PH value of the same, IPTG concentration is higher, the suicidal behavior, the more obvious; AHL is one of the important factors of bacterial suicidal behavior, at higher temperature, PH value under the condition of high AHL easy to decompose, reduced stability. So in same IPTG concentration, PH value at the same time, the training of the lower temperature, suicidal behavior, the more obvious; And IPTG concentration, same incubation temperature and same culture medium, the lower the PH, suicidal behavior, the more obvious. By comparing the experiment found that in the PH = 5.7 LB liquid medium add IPTG concentration of 1 mm, under 30 ℃ cultivate bacteria to suicide phenomenon is most obvious.
