陆地棉SIZ1基因的电子克隆及其分析

摘 要 :SUMO( small ubiquitin — related modifier)化修饰是细胞内蛋白质功能调节的重要方式之一,它是广泛存在于真核生物中高度保守的蛋白家族 ,在脊椎动物中有三个SUMO基因。SIZ1 在植物的抗寒性 、抗热性 、抗旱性 、磷酸饥饿反应 、抗病防御以及花期调控等生理过程中都发挥着重要作用。文章主要运用生物信息学的方法,利用拟南芥AtSIZ1基因在陆地棉中进行了电子克隆并没有检索到明确结果。利用拟南芥氨基酸或核苷酸序列在雷蒙德氏棉、亚洲棉数据库中比对检索获得序列,两者是串联顺向重复。利用雷蒙德氏棉、亚洲棉siz1a/b氨基酸或核苷酸序列在陆地棉ESTs数据库中比对检索获得序列9条序列,四个序列的比对结果完全一样。利用CAP3 Sequence Assembly Program,对陆地棉5’端和3’端ESTs分别进行组装获得了GhSIZ1基因5’端和3’端ESTs核酸序列。利用生物信息学的方法进一步对其DNA碱基序列与蛋白质氨基酸序列进行了分析,旨在为更好地研究基因SIZ1提供一定的理论资料。28531

In Silico Cloning and Analysis of GhSIZ1 Gene
Abstract:SUMO (small ubiquitin-related modifier) modification is one of the important ways of intracellular protein function adjustment, it is widely exists in eukaryotic highly conserved protein family, there are three type gene in vertebrates. SIZ1 in cold resistance, heat resistance, drought resistance, phosphate starvation response, antiviral defense, flowering regulation and physiological processes plays an important role in plant. This paper mainly using bioinformatics methods, the use of Arabidopsis thaliana AtSIZ1 gene to the upland cotton in electronic cloning is not retrieved clear results. Using Arabidopsis amino acid or nucleotide sequences in Raymond's cotton, cotton in Asia than in the database retrieval sequence, the two are in series consequent repetition. Use Raymond's cotton, cotton siz1a Asia/b amino acid or nucleotide sequences in upland cotton ESTs database than the retrieval for nine sequences, four sequence alignment results are exactly the same. Use CAP3 Sequence Assembly Program of upland cotton 5 'end and 3' end ESTs were assembled obtained GhSIZ1 gene 5 'end and 3' end ESTs nucleic acid Sequence. Using bioinformatics methods to further its DNA base sequence and protein amino acid sequence was analyzed, and aims at providing a better study SIZ1 gene with certain theoretical data.
