摘    要 本研究利用免疫荧光-激光共聚焦显微镜技术观察了中国石龙子精子发生过程中的微管分布特征,结合中国石龙子精子发生电镜图分析微管套在精子发生过程中的作用。实验结果如下:在精子发生时期Ⅰ时,细胞核的一侧有半圆形的凹陷,微管套暂未形成,微管分布不均,凹陷处的微管蛋白表达较强。在精子发生时期Ⅱ时,上一时期的核凹陷逐渐消失,核周围微管蛋白表达较强,微管在核周分布均匀,形成明显的微管套结构。在精子发生时期Ⅲ时,细胞核开始发生延长,核周围的微管套结构仍然紧紧包围核结构,与核的形状相同。在精子发生时期Ⅳ时,这个时期细胞核延长到最长,并在前端出现核尖,微管蛋白表达强烈,微管套紧紧围绕核周至精子发生过程结束。通过观察到的中国石龙子微管的动态变化,我们推测了有鳞目的分类地位,以及微管在精子发生过程中具有促进核发生形变的作用。89932

Abstract   In this study, immunofluorescence-based laser confocal microscope observes the microtubule distribution during spermiogenesis of Eumeces chinensis。 The experimental results are as follows: At the time of spermatogenesis period Ⅰ, there has a semi-circular depression in one side of the nucleus, microtubular sturcture-manchette hasn’t formation, the distribution of microtubules is nonuniform, the expression of tubulin was strong in the depression。 At the time of spermatogenesis period Ⅱ, nuclear depression of the last period gradually disappear, it has strong expression of tubulin around the nucleus, the distribution of microtubules is even, manehette was formed which surrounded the nucleus。 At the time of spermatogenesis period Ⅲ, nucleus has begin to lengthen, manchette were tightly attached to the nucleus periphery, and it has the same shape as the nucleus。 At the time of spermatogenesis period Ⅳ, nuclei extend to the longest in this period, nuclear prolongation appears at the front of the nucleus, it has strong expression of tubulin around the nucleus in this period, manchette were tightly attached to the nucleus periphery until the end of spermatogenesis。 Through the observation of the dynamic changes of microtubule during Eumeces Chinensis spermatogenesis, we speculate that the taxonomic position of the squamata, and the effect of microtubules in the process of spermatogenesis is promoting nuclear deformation。
