摘要桑树中的多种次生代谢产物,如黄酮、多糖和生物碱等,有降血糖、抗氧化、提高免疫力等作用,可以用作药物原料。植物细胞悬浮培养技术突破植物自身对于生长周期、地域和环境等因素的限制,可实现植物细胞代谢产物规模化生产的需求。本实验在成功建立桑悬浮细胞培养体系的前提下,研究了不同外源添加物(前体和诱导子)对桑悬浮细胞生长、黄酮含量、多糖含量以及1-DNJ含量积累的影响。实验结果表明:各外源物对桑悬浮细胞生长都表现出一定的抑制效果,不利于悬浮细胞生物量的积累;但对其次生物积累表现出不同的诱导作用。MJ、SNP、COR和苯丙氨酸对悬浮细胞黄酮的积累均有促进作用,以MJ诱导效果最佳;对于多糖的积累,仅有COR表现出明显的促进效果;MJ对细胞系中1-DNJ诱导作用显著,而SNP对1-DNJ的积累有抑制作用。各外源添加物对桑悬浮细胞生长以及次生代谢物的合成表现出不同的诱导作用,不同的诱导子刺激植物细胞启动不同的防御反应,产生不同的次生代谢产物。87266

Abstract Mulberry in a variety of secondary metabolites, such as flavonoids, polysaccharides and alkaloids, etc。, have fall blood sugar, anti-oxidation, improving immunity, etc, can be used as drugs raw materials。 Plant cell suspension culture technique can be used to help the plant to break through the limit of its own growth cycle, geographical and environmental factors, to achieve large-scale production of plant cell metabolites。 This experiment in successfully established under the premise of mulberry suspension cell culture system, studies on the different exogenous additives (precursor and inducer) for cell growth and accumulation of flavonoids, polysaccharide and 1-DNJ of mulberry suspension。 The results showed that the exogenous substances showed that all exogenous additives have inhibitory effect on cell growth, which is not conducive to cells suspended biomass accumulation; But for the second biological accumulation show different entrainment。 MJ, SNP, COR and phenylalanine for suspension cells are promoting effect, the accumulation of flavonoids in MJ induced effect is best。 For the accumulation of polysaccharide, only COR showed obvious promotion effect; MJ has induced effect significantly on 1-DNJ in cell lines, and SNP has inhibitory effect on 1-DNJ。 The exogenous additives for cell growth and secondary metabolites synthesis of mulberry suspension show different induction。 Elicitor stimulation of plant cells start different defense reaction, produce different secondary metabolites。
